| | MAO Inhibition Assays |
| Description: | The enzymatic reactions were carried out at pH 7.4 (K2HPO4/KH2PO4 100 mM, made isotonic with KCl) to a final volume of 500 µL. The reactions contained the different inhibitor concentrations spanning at least 3 orders of magnitude and the MAO-A/B mixed substrate kynuramine (45 µM for MAO-A and 30 µM for MAO-B). DMSO, as co-solvent (4%) was added to each reaction. The enzyme reactions were initiated with the addition of MAO-A or MAO-B (0.0075 mg protein/mL) and incubated for 20 min at 37 °C in a waterbath. After termination with the additionof 400 µL NaOH (2 N) and 1000 µL water, the concentrations of the MAO generated 4-hydroxyquinoline were measured by fluorescence spectrophotometry (excitation = 310 nM; emission = 400 nM). |
